A colon cancer tissue microarray containing 583 stage I-IV patients was stained for pan-cadherin, DAPI, and Met protein. Using novel automated curvature and intensity-based image analysis, epithelial membrane (red), cytoplasm (green) and nuclei (blue) were segmented. Stromal nuclei are shown in gray. The probability distribution of Met protein within each compartment was determined. A higher distribution of Met in the cytoplasm relative to the membrane was a significant predictor of survival in stage I and II patients.
